Why use estou here instead of sou?
Both verbs mean "to be", but estar is used for temporary things and physical locations. Since you are talking about where you are right now (in the bedroom), you use estou (I am). You would use sou (from the verb ser) for permanent traits, like your name or nationality.
Where is the word for "I" in this sentence?
The word for "I" is eu, but it is left out here. In Portuguese, the verb ending usually tells you exactly who the subject is. Because estou can only mean "I am", you do not need to say eu estou unless you want to add special emphasis.
What does no mean, and why is it used instead of em?
No translates to "in the". It is a mandatory contraction (combining of two words) of the preposition em (in) and the masculine word for "the", which is o. Because quarto (bedroom) is a masculine noun, we must combine em + o to make no.
