No, the "s" is completely silent. The word es is pronounced with a short "eh" sound, very similar to the "e" in the English word "pet".
Is es the only way to say "are"?

Strictly speaking, ici means "here" and là means "there". However, in everyday spoken French, people use là for both! If you want to say that you are present in a room, you will almost always say you are là.
