Use ไม่ใช่ to reject an identity, class, label, or equation: เขาไม่ใช่หมอ “he is not a doctor,” คำตอบไม่ใช่ข้อสาม “the answer is not number three.” Positive Thai may use เป็น for class membership and คือ for an identifying equation, but their ordinary negative often converges on ไม่ใช่.
English not appears after be. Thai does not build “be + not.” ไม่ใช่ comes before the noun phrase or identifying value being rejected. Actions and properties take basic ไม่ instead, while absence and non-presence conventionally use ไม่มี and ไม่อยู่.
Rejecting a class or role
When a positive clause classifies someone with เป็น, ไม่ใช่ normally rejects that classification.

The positive complements เภสัชกร and คนดูแล name classes or roles. No English article is translated before them. In the negative, ไม่ใช่ rejects the proposed membership as a whole.
Rejecting an identifying equation
Positive คือ identifies a value, person, cause, or answer. The negative is normally not ไม่คือ; use ไม่ใช่.

Choosing the negative by predicate type
Do not use ไม่ใช่ merely because English contains isn't or aren't. Thai property words are predicates without a copula, actions are verbs, มี expresses possession/existence, and อยู่ expresses location or presence.

ห้องนี้ไม่ใช่ใหญ่ is wrong because ใหญ่ is a property predicate. โรงแรมนี้ไม่ใช่มีที่จอดรถ is wrong because availability belongs to มี. The English surface form is not does not decide the Thai negative.
ไม่เป็น is real, but it says something else
Avoid the shortcut “ไม่เป็น is always wrong.” It is grammatical in other constructions. ว่ายน้ำไม่เป็น means “not know how to swim,” and แผลไม่เป็นอะไร means “the wound is nothing serious.” In a class or identity correction, however, ไม่ใช่ is the reliable choice.
ผมว่ายน้ำไม่เป็นครับ เลยไม่ลงสระลึก

This lexical contrast matters because English be encourages learners to treat every เป็น alike. Thai เป็น participates in class, state, and learned-skill constructions, and their negation differs by meaning.
Time, modality, and focus
ไม่ใช่ does not carry tense. Earlier or later reference comes from time expressions and discourse. Modal words can precede the whole negative equation: อาจไม่ใช่ “may not be,” คงไม่ใช่ “probably is not.”

The related pattern ไม่ใช่ว่า + clause means roughly “it is not that...” and negates a proposed explanation or inference. It is a clause-level extension, not the default way to negate a simple action.
Short replies depend on the question
To a confirmation such as คุณเป็นเจ้าของร้านใช่ไหมคะ, ไม่ใช่ครับ is a complete correction because the proposed identity is recoverable. To ไปไหมคะ “Are you going?”, reply ไม่ไปครับ, not ไม่ใช่ครับ.

The politeness particle keeps the short correction socially neutral. A bare ไม่ใช่ can be entirely grammatical, but in a service encounter it may sound abrupt depending on tone and relationship.
