Thai normally uses ไม่ใช่ to reject a nominal identity or classification: “not the manager,” “not a doctor,” “not mine,” or “not this meaning.” It serves as the ordinary negative counterpart across many positive sentences with เป็น or คือ, but it is not a negative form of English be for every predicate type.
A property takes ไม่ + property, a known entity's negative location takes ไม่อยู่, and absence or nonpossession takes ไม่มี. Choosing the negative therefore still requires you to identify the relation being denied.
Rejecting class membership and identity
The productive shape is subject/topic + ไม่ใช่ + noun phrase.
คุณวินไม่ใช่พนักงานประจำครับ

Khun Win isn't a permanent employee. (ordinary polite correction, male speaker; class membership denied)

In a positive characterization, Thai may use คุณวินเป็นพนักงานชั่วคราว. In an identifying equation, it may use ผู้จัดการคือคุณนิด. Their ordinary nominal negatives converge on ไม่ใช่. Thai does not mechanically create ไม่เป็น or ไม่คือ.
Ownership and attribution
ไม่ใช่ของ + owner rejects ownership. It differs from ไม่มี, which says a possessor lacks something.
กระเป๋าเดินทางใบสีเทาไม่ใช่ของฉันค่ะ

The gray suitcase isn't mine. (ordinary polite correction, female speaker; owner identification denied)

The first two sentences start from an identifiable item or idea and ask whose it is. The third starts from a possessor and denies that a suitcase lies in his domain. See ไม่มี for nonpossession.
ไม่ใช่ A แต่ B corrects the category
Thai commonly follows a rejected identity with the intended alternative.
นี่ไม่ใช่ใบเสร็จ แต่เป็นใบแจ้งหนี้ค่ะ
ni mai chai bai set tae pen bai chaeng ni kha
This isn't a receipt; it's an invoice. (ordinary polite correction, female speaker; category replaced)
ผู้รับผิดชอบไม่ใช่ฝ่ายบุคคล แต่เป็นฝ่ายบัญชีครับ

After the contrast, เป็น assigns a class or function, while คือ can highlight an exact identifying value. Everyday Thai often omits the second copula when the alternative is obvious: ไม่ใช่วันพุธ แต่วันพฤหัสบดี “not Wednesday, but Thursday.” Keeping it overt is useful when the structural contrast matters.

ไม่เป็น and ไม่ได้เป็น have other scopes
ไม่เป็น is real Thai, but not a universal nominal negative. It can deny becoming or entering a state, describe inability in a learned skill, or occur in lexical expressions. ไม่ได้เป็น often contrastively denies that a role actually holds.
ผมไม่ได้เป็นคนอนุมัติงบประมาณครับ

น้องชายผมขับรถไม่เป็นครับ

Compare plain เขาไม่ใช่เจ้าของร้าน, which simply classifies him as not the owner. ไม่ได้เป็น is especially natural when correcting a claim about what role someone had or has. These meanings are construction-specific; learners should not derive them by treating เป็น as an English auxiliary.
Rejecting an interpretation with ไม่ใช่ว่า
ไม่ใช่ว่า + clause rejects a conclusion or framing rather than one noun identity. It is common in careful conversation and explanatory writing.
ไม่ใช่ว่าผมไม่เห็นด้วย แต่ขออ่านรายละเอียดก่อนครับ
mai chai wa phom mai hen duai tae kho an railaiat kon khrap
It's not that I disagree; I'd just like to read the details first. (ordinary polite clarification, male speaker)

This frame negates an attributed proposition. It does not replace ordinary ไม่ inside every negative clause. In ผมไม่เห็นด้วย, ไม่ directly negates “agree”; in ไม่ใช่ว่าผมไม่เห็นด้วย, the speaker rejects that whole proposition as the correct explanation.
Answers and confirmation
Thai ใช่ can confirm an identity or proposition, while ไม่ใช่ rejects it. Short answers depend on what the question asks.
คุณเป็นผู้ประสานงานใช่ไหมคะ
khun pen phu prasan ngan chai mai kha
You're the coordinator, right? (ordinary polite confirmation question, female speaker)
ไม่ใช่ครับ ผมเป็นผู้ช่วยผู้ประสานงาน

For a property question such as ห้องนี้ร้อนไหม, the natural negative answer is ไม่ร้อน, not ไม่ใช่. Repeat or negate the predicate type actually under discussion.

Two staff members correct a delivery record:

1. Building negative identity with ไม่คือ
❌ คนที่โทรมาไม่คือคุณมาลีค่ะ
Incorrect—ordinary negative identification uses ไม่ใช่.
