Why is there a comma before друзі?

It’s the vocative case, plural. The noun друг (friend) uses друзі when you call or address multiple friends directly.

Pronounce it roughly as “dya-KOO-yu DROO-zi.” The stress falls on the second syllable of дякую (дя-ку́-ю) and on the first syllable of друзі (дру́-зі).

To be specific you can say подруги (female friends), vocative also подруги. So Дякую, подруги means “Thank you, [female] friends.” However, друзі is also often used for mixed or same-gender groups.
